1. PBI buffer provides the optimal salt concentration and pH for adsorption of high molecular weight DNA to the membrane. DNA adsorption to the silica-gel surface also requires a high
concentration of a "chaotropic agent" such as guanidine hydrochloride.
2.Buffer PE efficiently washes away contaminants. During the DNA adsorption step, unwanted contaminants such as primers, salts, enzymes, and unincorporated nucleotides do not bind to the silica membrane, but flow through the column.
3. Buffer QG has 2 functions, 1). solubilize the agarose gel slice; 2). provide the appropriate conditions for binding of DNA to the silica membrane, pH<7.5.

以DNA纯化为例，第1个Buffer的作用是裂解细胞，第2个Buffer 的作用是去蛋白，第3个Buffer 的作用是去盐，最后一个Buffer 的作用是溶解DNA。
胶回收时的QG buffer除了溶胶还有调节PH值的作用，在特定的PH值下，DNA会吸附在膜上，然后用水或者TE Buffer溶解DNA
